John Dillinger Josh Perkins On June 22, 1903 a man named John Dillinger was born. He grew up in the Oak Hill Section of Indianapolis. When John was three years old his mother died, and when his fatehr remarried six years later, John resented hes stepmother. When John was a teenager he was frequently in trouble. He finally quit school and got a job in a machine shop in Indianapolis. He was very intelligent and a good worker, but he soon got bored and often stayed out all night. His father began to think that the city was corrupting his son, so he sold his property in Indianapolis and moved his family to a farm near Mooresville, Indiana. John reacted no better to rural life than he had to that in the city and soon began to run wild again. At the age of 21 he attempted his first robbery, robbing a grocery store, in his home town. He was caught and imprisoned for nine years until 1933. Soon after he was released, Dillinger robbed a bank in Bluffton, Ohio and was arrested by the Dayton police. He was put in Lima county jail to wait for his trial. The Lima police found a document on John which seemed to be a plan for a prison break, but he denied everything. Four days later, using the same plans, eight of Dillinger's friends escaped from the Indiana State Prison, using shotguns and rifles which had been smuggled into their cells. During their escape, they killed two guards. On October 12, three of the escaped prisoners and a parolee from the same prison showed up at the Lima jail where Dillinger was. They told the sheriff that they had come to return Dillinger to the Indiana State Prison for violation of his parole. When the sheriff asked to see their credentials, one of the men pulled a gun, shot the sheriff and beat him into unconsciousness. They took the keys, freed Dillinger, locked the sheriff's wife and a deputy in the cell, and left. Leaving the sheriff to die on the floor. These four mens fingerprint cards were pulled, indicating that they were wanted. Meanwhile, Dillinger and his gang pulled several bank robberies. They also stole several machine guns, rifles, and revolvers, a quantity of ammunition, and several bulletproof vests. By mid-1934, Dillinger had been involveed in at least ten bank robberies in Indiana, Ohio, Wisconsin, and South Dakota. He was hiding in Chicago with a woman by the name of Anna Sage. This acquaintace was said to be the one who told on John. She told federal agents she would be wearing a red dress when she and a girl friend accompanied Dillinger to the theater. Sage became famous as the "lady in red." Agents were waiting outside to arrest him. Dillinger grabbed a pistol from his trouser pocket and attempted to flee, the three agents opened fire. Dillinger was pronounced dead at 10:50 p.m. at Alexian Brother's Hospital. John Herbert Dillinger was killed July 22, 1934 in front of a Chicag movie theater. He was shot four times. Two shots grazed his face, one shot pertruded into is arm ,and the fourth shot entered the back of his head, exiting underneath his right eye. John was 31 years old when he died. He had been at the top of the Federal Bureau of Investigation's "Ten Most Wanted" list for a month before he died. How to Find Your Goal Score So how do you find out what makes for good ACT scores for the colleges you’re interested in? In this section, we’ll walk you through a quick five-step process for figuring out the best goal score for you. Step 1: Download This Worksheet To work through the following steps, we'll be filling out a worksheet for all the schools you're planning to apply to. Click here to download it, or click the image below. I recommend you print it out so that you can write on paper and keep it next to your work space. Step 2: Fill in the Schools You Want to Get Into on the Left Fill in all the schools you're sure you want to apply to already in the leftmost column. If you don't know what schools you're aiming for yet, feel free to use ones that have been suggested to you, or schools that your friends are interested in. I recommend that you take the time to research schools first, though, so that you have a realistic target score.Because your goal score should be targeted to the schools you are actually interested in applying to, the more accurate of an idea you have of the schools you want to apply to, the more accurate your goal score will be. Step 3: For Each School, Google "[Name of School] PrepScholar ACT" For example, if I'm interested in U Alabama, I'll do the following search: Click on the the ACT and GPA post (or the Admission Requirements post, they’ll both have the information) and scroll down to find the 25th and 75th percentile composite ACT scores for admitted students. For University of Alabama, the 25th percentile score is 23. As a quick reminder, the 25th percentile means that 25% of admits have a score at or below that number. So 23 would be a below average score for admitted students for U Alabama. The 75th percentile score for University of Alabama is 32. That means that students with that composite score scored better than 75% of all the other admits. So scoring at that level or above puts you in the top quarter of admits score-wise- a very competitive score! In summary, the 25th/75th percentile range describes the scores of the middle 50% of all students admitted to a particular school. If you score at the 75th percentile for any school, you have a great chance at getting in (assuming your other credentials are appropriate for the school). If you're at the 25th percentile, you'll need to have a particularly strong application to boost your odds of getting in. For each school on your list, google the PrepScholar ACT score information and write down the 25th and 75th percentile scores in the appropriate row for that school on your goal score sheet. Step 4: Calculate Your Final ACT Target Score To calculate your target ACT goal score, look at the 75th percentile column. Find the highest score in that column. That’s your composite score goal. If you score at the 75th percentile level for the most competitive school on your list, you’ll be competitive at all your schools for test scores. Another advantage of choosing a high goal score is that if you end up falling 1-2 points short, it’s not a huge deal because you’ll still be competitive for most of your schools. You might be thinking- hey, wait! Why did I fill out that entire sheet if I was just going to pick the highest 75th percentile score? Well, the advantage of filling out all that information is that you now have it handy as a reference. You’ll be able to check your ACT score against all your schools of interest as soon as you have it! Step 5: Share Your Target Score As a last step, I suggest that you do two things with your score target: Share it with your parents. This will be a helpful conversation around your personal goals and how you want to achieve your target ACT score. Plus, they can help keep you accountable throughout the preparation process! Tape it to your wall. This will keep your goal in mind, which will help keep you motivated to prepare. Keep your goals in sight! If you take the test and you get lower than your goal score, what should you do? Don’t panic; you have a few options. We’ll go over them here and help you figure out when you should consider them. Strategy 1: Retake the Test If you have the time to prepare for the test and re-take it, this is probably your best strategy if you are really set on all your schools. (Unless you were only 1 or maybe 2 points under, in which case it might actually be a poor use of time toretake the test- see strategy #2). You’ll want to be sure to do targeted prep of your weaknesses to have the best chance of actually improving your score. You should also make sure you actually prepare for enough hours to make the meaningful difference in score that you need. Here are our rough estimates for how many hours of prep it will take for you to improve your composite score by a certain amount: 0-1 ACT Composite Point Improvement: 10 hours 1-2 ACT Point Improvement: 20 hours 2-4 ACT Point Improvement: 40 hours 4-6 ACT Point Improvement: 80 hours 6-9 ACT Point Improvement: 150 hours+ Strategy 2: Don’t Worry About It If you only missed your goal score by 1-2 points, depending on the schools you’re applying to, you might not need to do anything. Let’s say you were going for a 35, but you got a 34. You could retake the test, but you don’t necessarily have to. If your 34 still puts you towards the top of the 25th-75th percentile range for your schools, it might make more sense to use the time and energy you would spend prepping for and retaking the test on other parts of your application. However, if you were more than two points short of your goal score, you should consider strategies 1 or 3. And if you’re applying to very selective schools, even two points might make a retake worth it. Strategy 3: Adjust Your List of Schools If you are 3+ points short of your goal score and you don’t have time to retake the test, your best strategy is to adjust your list of schools. While you can (and should) still apply to your dream schools as reach schools, you’ll need to make sure that you have enough strong match and safety schools for your scores. Let’s say you were going for a 32, but you got a 28. You may have had Boston University(middle 50% 29-32)as a match, but now it’s more of a reach. And maybe you had Hofstra University (middle 50% 24-29) as a safety school but now it’s closer to a match. So add some safety schools for your score, like SUNY Albany (middle 50% 22-26) and Pace University (middle 50% 22-27).You can see more on choosing appropriate safety, match, and reach schools here. Biography of Ralph Waldo Emerson Ralph Waldo Emerson was one of the most influential Americans of the 19th century. His writings played a major role in the development of American literature, and his thought impacted political leaders as well as countless ordinary people. Emerson, born into a family of ministers, became known as an unorthodox and controversiall thinker in the late 1830s. His writing and public persona would cast a long shadow over American letters, as he influenced such major American writers as Walt Whitman and Henry David Thoreau. Early Life of Ralph Waldo Emerson Ralph Waldo Emerson was born May 25, 1803. His father was a prominent Boston minister. And though his father died when Emerson was eight years old, Emersons family managed to send him to Boston Latin School and Harvard College. After graduating from Harvard he taught school with his older brother for a time, and eventually decided to become a Unitarian minister. He became the junior pastor at a noted Boston institution, Second Church. Personal Crisis Emerson’s personal life appeared promising, as he fell in love and married Ellen Tucker in 1829. His happiness was short-lived, however, as his young wife died less than two years later. Emerson was emotionally devastated. As his wife was from a wealthy family, Emerson received an inheritance which helped sustain him for the rest of his life. The death of his wife and his plunge into misery led Emerson to have severe doubts about his religious beliefs. He became increasingly disillusioned with the ministry over the next several years and he resigned from his position at the church. He spent most of 1833 touring Europe. In Britain Emerson met with prominent writers, including Thomas Carlyle, which whom he began a lifelong friendship. Emerson Began to Publish and Speak in Public After returning to America, Emerson began to express his changing ideas in written essays. His essay â€Å"Nature,† published in 1836, was noteworthy. It is often cited as the place where central ideas of Transcendentalism were expressed. In the late 1830s Emerson began to make a living as a public speaker. At that time in America, crowds would pay to hear people discuss current events or philosophical topics, and Emerson was soon a popular orator in New England. Over the course of his life his speaking fees would be a major portion of his income. The Transcendentalist Movement Because Emerson is so closely linked to the Transcendentalists, it is often believed that he was the founder of Transcendentalism. He was not, as other New England thinkers and writers actually came together, calling themselves Transcendentalists, in the years before he published â€Å"Nature.† Yet Emerson’s prominence, and his growing public profile, made him the most famous of the Transcendentalist writers. Emerson Broke with Tradition In 1837, a class at Harvard Divinity School invited Emerson to speak. He delivered an address titled â€Å"The American Scholar† which was well-received. It was hailed as â€Å"our intellectual Declaration of Independence† by Oliver Wendell Holmes, a student who would go on to be a prominent essayist. The following year the graduating class at the Divinity School invited Emerson to give the commencement address. Emerson, speaking to a fairly small group of people on July 15, 1838, ignited a huge controversy. He delivered an address advocating Transcendentalist ideas such as love of nature and self-reliance. The faculty and clergy considered Emerson’s address to be somewhat radical and a calculated insult. He was not invited back to speak at Harvard for decades. Emerson Was Known as The Sage of Concord Emerson married his second wife, Lidian, in 1835, and they settled in Concord, Massachusetts. In Concord Emerson found a peaceful place to live and write, and a literary community sprang up around him. Other writers associated with Concord in the 1840s included Nathaniel Hawthorne, Henry David Thoreau, and Margaret Fuller. Emerson was sometimes referred to in the newspapers as The Sage of Concord. Ralph Waldo Emerson Was a Literary Influence Emerson published his first book of essay in 1841, and published a second volume in 1844. He continued speaking far and wide, and it’s known that in 1842 he gave an address titled â€Å"The Poet† in New York City. One of the audience members was a young newspaper reporter, Walt Whitman. The future poet was greatly inspired by Emerson’s words. In 1855, when Whitman published his classic book Leaves of Grass, he sent a copy to Emerson, who responded with a warm letter praising Whitman’s poetry. This endorsement from Emerson helped launched Whitman’s career as a poet. Emerson also exerted a major influence over Henry David Thoreau, who was a young Harvard graduate and schoolteacher when Emerson met him in Concord. Emerson sometimes employed Thoreau as a handyman and gardener, and encouraged his young friend to write. Thoreau lived for two years in a cabin he built on a plot of land owned by Emerson, and wrote his classic book, Walden, based on the experience. Involvement in Social Causes Emerson was known for his lofty ideas, but he was also known to get involved in specific social causes. The most notable cause Emerson supported was the abolitionist movement. Emerson spoke out against slavery for years, and even helped runaway slaves get to Canada via the Underground Railroad. Emerson also praised John Brown, the fanatical abolitionist who many perceived as a violent madman. Though Emerson had been fairly apolitical, the conflict over slavery led him to the new Republican Party, and in the election of 1860 he voted for Abraham Lincoln. When Lincoln signed the Emancipation Proclamation Emerson hailed it as a great day for the United States. Emerson was deeply affected by Lincolns assassination, and considered him a martyr. Emersons Later Years After the Civil War, Emerson continued to travel and give lectures based on his many essays. In California he befriended naturalist John Muir, whom he met in Yosemite Valley. But by the 1870s his health was beginning to fail. He died in Concord on April 27, 1882. He was nearly 79 years old. His death was front-page news. The New York Times published a lengthy obituary of Emerson on the front page. It is impossible to learn about American literature in the 19th century without encountering Ralph Waldo Emerson. His influence was profound, and his essays, especially classics such as Self-Reliance, are still read and discussed more than 160 years after their publication. What can be done to prevent prejudice from occurring? Preventing prejudice is a challenge, although as individuals we can change the way our younger generations see other people, but to prevent it as a whole is impossible. Lesson Before Dying The Right to Be Free In the novel A Lesson Before Dying, although Grant is an educated black man in the era of a racist society he has struggles greater than most men of his decent. I feel sorry for him because of his limitations, even though I view him as a coward. He cannot break free of his background and family. The three main female characters in the novel, Tante Lou, Miss Emma, and Vivian, restrict and limit Grant's choices. Grant realizes that freedom means leaving his small town and creating a new life, yet each woman holds a chain that keeps him from his destiny and the right to be free. First, Tante Lou, his aunt holds Grant from his dreams by refusing to let him go his own way. Tante Lou wants Grant to stay at home with her and take care of her. But, in the time the novel takes place, it wasn't common for young men stay with their elders and help out, especially when Grant has a college degree and can accomplish so much. Tante Lou took advantage of this in any means necess ary, using the fact that Grant is family and in essence owes it to her to stay and help out. We can see this on page 14 when Tante Lou forces Grant to talk to Mr. Henri about seeing Jefferson. You are going up there with us Grant, or you will not sleep in this house tonight. Human and Disturbed Young Member Essay

I do so loathe writing essays, especially a descriptive one, but for the grade I’ll just have do it. The topic I chose to write about as you read in the title is myself. To describe how I am and the way I am should prove an easy task. To explain why I am this way is a different story. Furious, despondent and hopeless I have felt, and have endured feelings similar for a rather long period of time. In the end, like it or not this essay will be completed. For starters, emotionally I am not considered a very cheery or content individual. My mood for half of an average day is cantankerous, or rather irritable. The other half is I would say filled with cheerless and heartbreaking misery. On the rare days that I am in high spirits, habitually my short ill temper will devastate my day along with anyone else’s if I am amongst others. I don’t seem to make nor keep friends very well due to my offensive attitude. I am moderately unpleasant to be around and somewhat unkind to those around me. One must realize that to be friends with me is reasonably difficult chore. Next, following my emotions is my state of mind. I regret to announce that the stability of my mentality is delicate and unreliable. Not to indicate that I am mentally ailing or handicapped nevertheless I do not speak of my brain literally for that is a stronger and different matter. I as a person am fragile and feeble although my faà §ade shows to the contrary. Pushed too hard and reluctantly I will not last for too long. I lack determination and willpower when it comes to my mind and aiding in its recuperation. I’d rather lock it all up inside and bury it deep down never to resurface again until it just detonates. Through my neglect, I have fractured my mind and made myself a very disturbed young member of society. Apart from all the consolers and therapists, my family tries to show their love and support. Though it doesn’t help much, I just put on a smile and present them with what they want to see. My brain on the other hand is stronger and more resilient than my mind or will. I once thought of myself as naà ¯ve and unaware of society as a whole growing up in a small town alone. People and their actions or what they said, even how they looked, was mysterious and unknown to me. Un-relatable I thought the human race to be. Moving to the very overpopulated city, I now realize that not only was I not naive, but that I knew more than I cared to know. All the same, all predictable, the only difference is that here I fit in because no one cares to notice. When you live in a town where everyone notices, one feels very inexperienced, childish almost under the watchful eyes of others. Growing up alone, one is obligated to solitude and when you are watching everyone else, observations are what one’s childhood is based on. From just examining a setting or situation, even people, I can analyze it and bring myself to hypothesis that ends up being fairly close to the actual occurrence . Experience wise, I am somewhat an aware young citizen of the community. Living my life has been no easy job although I’m sure the rest of the world feels the same. My skills learned throughout life are quite a handful. Book smarts was one of my highest assets and at one point based my life around it. I grew up with nothing but my books and thought they were everything I need and would ever need. In this world it’s better to fill your head with knowledge, for when all materialistic items have passed your brain still remains. To this day however I play the part of an idiot, truth be told it makes things far less complicated when they think you are an incompetent moron. Another motive behind my opinion that I was naà ¯ve was that I generally did not get along with kids my age. No matter how hard I tried, relating to them was a vast issue. I believed that I was too immature for them to relate to. Only came to discover that I was the one too mature for them. My circle of friends is a very small one and the people it consists of are all over the age of about twenty. For a reason unpronounced to me I get along famously hand in hand with them. Perhaps it is that their maturity is vastly to the contrary of any stubborn adolescent. I couldn’t hold a conversation with a fellow teen for any extended amount of time, even if I genuinely sought to. My motto now is that if they want to talk to me, they will get up and try. If they want to befriend me, they will make the effort. No longer will I strain myself trying to talk to someone who doesn’t give me the time of day nor upset myself when I’m alone again. In the end, along with a descriptive essay in full, I have expressed to thee my thoughts and my opinions. I am a seriously distressed human being with pretty severe mood swings. Incorporate my rigid attitude, weak state of mind, hidden knowledge and the combination is thick with incompatible components. Collaborate my experiences with the fact that I am now a stronger and better person through previous anguish, and I you have a very strong and resilient victim. Being accustom to seclusion was what made me such an awful person. I admit my mental health is perchance unstable and my maturity prohibits me to make teen friends on a substantial level. The economists, government representatives and many politicians said that Hungary’s economy is growing. Growing half times as it should be. In reality and if economics can only speak, Hungary was in big trouble and in the verge of economic collapse last year. The economic crisis of Hungary led the resignation of Prime Minister Ferenc Gyurcsany. Now, Hungary’s population is looking for the proper replacement for their Prime Minister. And they hope that whoever would be their Prime Minister will help them regain their economic stability. As economic crisis still exists in the country, many believed that the major factor that would determine the economic stability is its policies regarding their pensioners. Hungary’s total population of pensioners totaled for about 3 million which is about one-third of the whole population. Aside from supporting these pensioners, the government also pays for special benefits to accident victims, military and police veterans. The problem boils down to the public spending and finances. Pension is a heavy obligation that Hungary’s government is shouldering because of their policy regarding pensioners. Reforming the policy is not an option for the Socialist party of Gyurcsany because they protect the Socialist voting percentage which is mainly composed of retirees. Solutions should be done and we should try anything possible that will be in great help for the recovery of the nation. Possible solution is to cut the pension received by the retirees into much lesser amount. This would definitely be a great help in the recovery. One another option is to limit the pensions to those in proper age of retirees and not to those who were only taken their opportunity of having their pension because of undersized accidents. In the end, all should be working for the progress and recovery of this nation, not only the officials but also the people that are the ones greatly affected by the economic crisis. Any solutions can be beneficial and should be tried.

Techniques for the Identification of Unknown Microorganisms Identification of Unknown Microorganisms For years and years, we have been trying to classify and understand microorganisms, and in doing so, we learned of certain techniques to identify them based on what they look like, where they live, and what they eat. We used this knowledge to derive simple tests to run, and we used a key to narrow down our choices, all in an effort to identify a microorganism. Beginning in the earliest labs with the earliest microbiologists, the most common way to identify an organism is to look at it and look at what it does. In microbiology of bacteria, is it round (cocci) or rod-shaped (bacilli)? Does it stain purple (gram +) or red (gram -)? Does it use lactose as food, while creating an acidic product that can be observed? These tests based on the physiology/morphology and biochemistry fall under what is known as classic identification  [1]. Classic techniques focus on the ability of a bacterium to grow and reproduce in certain conditions, such as the fact that bile salts can interact unfavora bly with the peptidoglycan cell walls of gram positive bacteria, therefore selecting against their growth. Most tests are used to specifically select for or differentiate between the species, this in conjunction with a dichotomous key helps narrow down the choices until one is satisfactory. In contrast, modern methods of identification are based on the technological advances of recent years, namely, polymerase chain reaction (PCR) and genetics. The use of PCR to identify bacteria is rooted in the study of genetic coding, in particular, RNA. Every bacterium has strands of RNA that is used to bind the mRNA for translation: the 16s ribosomal RNA (rRNA)  [2]. This sequence is conserved in all bacteria and can be used to identify species after amplification through the PCR process. Since every species has a different genetic make-up, it is simply a matter of time for identification. But for most purposes, getting samples to a PCR and running them takes quite some effort, and a lot of m oney, thus the simpler and more efficient method is the classical testing. At one point or another, we all have gotten the sore throat and cough. These symptoms are sometimes caused by microbes in the pharynx that shouldnt be there. Bacteria such as Streptococcus pneumonia, Haemophilus influenza, and Moraxella catarrhalis can infect the area and cause this sore feeling. A more serious offender, Streptococcus pyogenes, causes what is commonly known as strep throat. These bacteria make the pharynx sore by harvesting erythrocytes and breaking them down as sources of food, in turn, destroying the lining and causing the all familiar sore feeling1. A simple test on a blood supplemented agar helps reveal what source of pathogen is causing the sickness. The agar is inoculated and incubated until growth is seen, and the plate will show a pattern of hemolysis if caused by one of the bacterial pathogens. Beta hemolysis is the ability to fully metabolize the cell and hemoglobin and will produce a clear halo around the colonies. Alpha hemolysis is the partial breakdowns of hemoglobin into billiverdin, making the halo appear yellow/green. Gamma hemolysis is the absence of halo, as it means the organism cannot lyse erythrocytes1. Procedures: Respiratory swab: Take a sterile cotton swab and swab the tonsilary region Inoculate blood agar by rolling the cotton tip in a streak plate method After the initial line, discard the swab in biohazard bin, and continue with a flamed inoculation loop Second swab: Take a sterile cotton swab and swab the tonsilary region Place the swab in a test tube Dilute to 10-3 and 10-4 Make a pour plate with nutrient agar Unknown identification: To start to identify any unknown organism by classical methods, it is necessary to create an appropriate dichotomous key to accompany the tests. The first step to any key is the gram stain; this essentially splits the number of possible organisms in half, gram positive and negative. Further tests based on morphology and biochemistry will select and differentiate between common characteristics of species until one organism stands out. At the start of every lab day, fresh streak plates and broths are prepared to keep the organism isolated and fresh. Take a single colony and prepare streak plates, one for 37 °C and one for 25 °C Prepare broths aseptically in similar fashion Dichotomous key: The dichotomous key was prepared using only the organisms on the list provided by Dr. Maxwell. Gram stain: The gram stain separates bacteria into two main groups, gram positive and gram negative. The peptidoglycan cell walls of gram positive organisms will absorb the crystal violet stain and appear purple under microscope inspection. The gram negative bacteria lose their outer membrane during the decolorization step and will absorb the safranin stain, appearing red. Prepare specimen by taking an isolated colony and heat fixing on a slide Apply crystal violet stain for 60 sec Wash with water Apply iodine fixer for 45 sec Wash with water Rinse with decolorizer for exactly 8-10 sec Wash with water Apply safranin for 60 sec If gram positive, the next step in my dichotomous key is to differentiate based on morphology: is it a cocci or bacilli? This step will split the organisms by shape, essentially cutting the number in half. Using the gram stain slide, look at the shape of the specimen under 100x oil immersion Bacilli are rod-shaped cocci are round If bacilli, run the catalase test. The catalase test will reveal if the organism has catalase, an enzyme used to breakdown harmful oxidizing agents such as hydrogen peroxide, which could be made as a result of metabolism. A drop of hydrogen peroxide is added to the specimen placed on a slide. A positive result will cause bubbles to form, indicating presence of catalase. Put a colony on a slide Add hydrogen peroxide Observe bubbles/no bubbles If catalase negative, as in no bubbles formed, the organism is Lactobacillus acidophilus. The bacteria are part of healthy normal flora and aids in digestion  [3]  . If catalase positive, the endospore stain must be used for further classification. The endospore is a tough spore formed by some bacteria during harsh environments. The spore will be dyed green in contrast to a cell dyed red. Prepare a specimen by heat fixing on slide Cover the slide with bibulous paper Add methylene blue Place slide on steamer over a Bunsen burner Steam for 7-10 minutes, applying additional methylene blue to prevent the paper from drying out Remove slide and paper Cool for no more than 30 seconds and rinse Counterstain with safranin for 1 minute Rinse and blot dry If endospore positive, the bacteria is Bacillus subtilis. The organism is found readily in the soil and can form a tough endospore when conditions become unfavorable. Bacillus is also a great model organism for gram positive studies  [4]  . If endospore negative, the organism is Corynebacterium diphtheriae. This bacterium is the cause of diphtheria, a respiratory infection. If the organism observed is cocci, run the catalase test. The catalase test will reveal if the organism has catalase, an enzyme used to breakdown harmful oxidizing agents such as hydrogen peroxide, which could be made as a result of metabolism. A drop of hydrogen peroxide is added to the specimen placed on a slide. A positive result will cause bubbles to form, indicating presence of catalase. Put a colony on a slide Add hydrogen peroxide Observe bubbles/no bubbles If catalase negative, the organism is Enterococcus faecalis. It is commonly found in the digestive tract of humans and is considered part of the normal flora. It does not produce catalase. If catalase positive, the use of mannitol salt agar is used to differentiate between the staphylococcus and micrococcus bacteria. Mannitol salt agar is used to select for staphylococcus. Staphylococcus aureus will produce a yellow halo. Inoculate the MSA plate with a isolated colony Incubate at 37 °C for 48 hours Observe growth If a yellow colored halo around the colony is observed, it is either S. aureus white or S. aureus gold. The way to differentiate between the two is the color of the colony itself, which respectively is, white or golden (yellow). S. aureus is part of the normal flora and what is the usually cause of infections and pimples. If the MSA plate shows no yellow halo, then it is either Micrococcus roseus or Staphylococcus epidermis. The way to differentiate between them is the color of the colony itself. Micrococcus will appear red in color due to a pigment it secretes  [5]  , while S. epidermis will be a white color. Both are normal skin flora. If the organism is gram negative, the first test to run is the lactose supplement phenol red broth. The purpose of this test is to indicate whether or not the organism can use lactose as a food source. Fermenting lactose will produce an acidic waste and cause the phenol red to change color. A positive test will turn yellow. Inoculate a tube with an isolated colony Incubate at 37 °C for 48 hours Observe color change If the organism cannot utilize lactose, then perhaps it can use glucose as a food source. The glucose phenol red broth tests for a similar metabolic process as the lactose one. Inoculate a tube with an isolated colony Incubate at 37o C for 48 hours Observe color change If the bacteria are glucose negative, then it is either Pseudomonas (Burkeholderia) cepacia or Pseudomonas aeruginosa. The two can be differentiated by the pigment of the colonies. P. cepacia will appear a light purple color while P. aeruginosa will be white, and also produce a green pigment known as pyocyanin  [6]  . If glucose positive, a citrate slant must be prepared. The citrate slant tests for the ability to use citrate as a carbon source. A positive result will turn the green slant blue. Inoculate the slant with an isolated colony Incubate Observe color If citrate negative, the bacteria is Proteus vulgaris. It is a bacterium that normally inhabits the intestines of humans and in certain cases can cause infections, albeit in most cases of immunocompromised individuals. If citrate positive, the organism is Serratia marcescens. It also produces a red pigment that can be used to identify it. The bacteria are found throughout environments and can be observed upon bathtubs that dont get washed often  [7]  . If the organism can ferment lactose, then the citrate test is prepared. The citrate slant tests for the ability to use citrate as a carbon source. A positive result will turn the green slant blue. Inoculate the slant with an isolated colony Incubate Observe color If citrate negative, the bacteria is E. coli. This is one of the most used model organisms for gram negative studies in labs. The rod shaped bacteria can cause some food poisoning if ingested. But the best use of it is in the labs, as it is easy to care for, replicates fast, and genetically simple, allowing for genetic research  [8]  . If citrate positive, the bacteria could be either Enterococcus or Citrobacter. The test to differentiate between the two is a Methyl red à ¢Ãƒ ¢Ã¢â‚¬Å¡Ã‚ ¬ Voges Proskauer media. The MR-VP is used to differentiate bacteria based on the ability to hydrolyze dextrose and create stable organic acids during fermentation. The MR-VP test combines the two separate tests. The positive result for the methyl red test is a red color when the methyl red indicator is added to the media. The positive result for the Voges Proskauer test is a red color. Inoculate two MR-VPs Incubate Add the MR reagent to one tube and observe any color change every 10 mins Look for any color change for the VP test before addition of reagent If MR-VP positive, the organism is Enterobacter. E. aerogenes can cause infections and is considered pathogenic; however it still does exist as part of the normal flora of the human intestines. If MR-VP negative, the organism is Citrobacter. They are ubiquitous in the environment and also inhabit the intestines. Results: Respiratory swabs: The streak plate to identify organisms with hemolytic abilities showed growth of many different bacteria morphologies. Green and white colonies were observed to be growing on top of the plate. The media remained red throughout; no indication of any white spots. The pour plates for were both too numerous to count. Unknown organisms: The organisms were isolated using a streak plate. Two isolations were obtained at two temperatures, 25 ­o C and 37o C. the plate at room temperature showed red pigmented colonies (B), while the body temperature plate showed thick white colonies (A). A third unknown was given pre-inoculated (C). Three biochemical tests and two morphological tests were used to identify the first unknown organism after isolation. A gram stain revealed that the organism was gram positive; being violet under the 100x oil immersion lens. Also while under the microscope, it was revealed that organism A was cocci. A catalase test was done and organism A was determined to be catalase positive, producing bubbles after addition of hydrogen peroxide. The unknown bacteria were inoculated on a mannitol salt agar, which showed the halo, a positive result. To identify between the two staphylococci aureus, the pigment of the colonies was used: white. A gram stain revealed that organism B was gram negative; red under the 100x oil immersion lens. A lactose and glucose tube was inoculated in the same period to identify the ability to utilize those sugars. The organism was not able to metabolize lactose and was able to use glucose. A citrate slant was inoculated, after 48 hours the butt and slant of the citrate slant was turned blue; positive result. The third unknown was tested to be gram positive. A catalase test came out negative, producing no bubbles. Conclusions: Respiratory swab: Because no halo or clearing of any kind was observed in the media around the colonies, it can be safely said that none of the organisms in our throat swabs contained bacteria capable of hemolysis. This lack of hemolytic ability is known has gamma hemolysis. The pour plates were TNTC. This is probably because the dilutions performed were not enough; too many organisms were taken per tonsil swab. At the time, both participants were mildly sick and had seasonal allergies, possibly contributing to the amount of organisms per swab. Unknown organisms: Three unknown organisms were presented for identification. Organism A was successfully identified as Staphylococcus aureus white. What lead to that identification is that S. aureus is a gram positive cocci that produces catalase and is able to use mannitol and grow on the mannitol salt agar and produce the halo; it was determined that the color of the colonies was white as opposed to gold. S. aureus is a common bacteria found on the skin and part of human normal flora. The bacteria can cause minor infections of the skin and can sometimes cause serious diseases and infection if it reaches the systemic circuit  [i]  . Organism B was identified as Serratia marcescens. It is a gram negative rod that can ferment glucose and citrate, but not lactose. It is a common environmental bacterium that can sometimes cause infections in skin and the urinary tract. S. marcescens is part of the family enterobacteriaceae and produces a red pigment that can be found on bathtubs and tiles, feeding on soap residues  [ii]  . Organism C was identified to be Lactobacillus acidophilus. It is a gram positive rod that does not produce any catalase for hydrogen peroxide breakdown. The bacteria are part of Lactobacillus which ferments sugars into lactic acids. This bacterium is used often for the production of many dairy products such as cheese and yogurt. L. acidophilus is part of normal human flora and can be found on the skin or gut. Sometimes it is used as a probiotic supplement. It also prevents Candida from overgrowing in the female vagina  [iii]  . All three organisms were identified successfully with the use of classic techniques.

A Destructive Society Exposed in Maggie In Maggie, Stephen Crane deals with poverty and vice, not out of curiosity or to promote debauchery but as a defiant statement voicing the life in slums. Drawing on personal experience, he described the rough and treacherous environment that persisted in the inner-city. By focusing on the Johnsons, Crane personalizes a large tragedy that affected and reflected American society as a whole. His creation of Maggie was to symbolize a person unscathed by their physical environment. Through Jimmie he attempted to portray a child raised without guidance who turned into his abusive, drunk father. Crane plays Jimmie and Maggie off of each other as opposites. The Mother and Father are depicted as failed drunken hypocrites and poor role models. Crane skillfully characterizes and stereotypes the personalities in Maggie to illustrate the influence of environment and the wretched conditions in slums. Maggie "blossomed in a mud puddle" and represented purity in a corrupt world. When she gets together with Pete she attempted to get out of the world she despised, but instead remained in the slum, unable to escape. Although she is repeatedly abused, Maggie continually picks up the remnants of her life despite being "in a worn and sorry state." Jimmie is seen both in a good light, like his sister, as well as an evil and cruel person. In the beginning of the story, he is portrayed as the "little champion" of Rum Alley. However, that description merely cloaked the brutal fight that he was engaged in and the beating he later gave his sister. Later in the story, Jimmie buys some beer for an old leathery woman, but it is taken by his father. Jimmie protests in the name of justice but is not successful. The crude and abusive relationship with his father severely cripples his chances to become a benevolent adult. Instilled with poor values he did not see the world as good and bad but rather bad and worse. When he "studied human nature in the gutter, and found it no worse than he thought he had reason to believe it" he expressed his pessimistic and cynical attitude towards the world. The Johnson's mother is typical of a drinking, abusive, and careless mother. She stood for a hypocritical, industrializing society that was neglecting its children. When Jimmie tries to take his mother home when she has been kicked out of a bar "she raise[s] her arm and whirl[s] her great fist at her son's face.
